RESUMEN

Knowledge of genetic diversity among genotypes and relationships among elite lines is of great importance for the development of breeding programs. Therefore, the objective of this study was to evaluate genetic variability based on the morphoagronomic and molecular characterization of 18 elite popcorn (Zea mays var. everta) lines to be used by Universidade Estadual de Maringá breeding programs. We used 31 microsatellite primers (widely distributed in the genome), and 16 morphological descriptors (including the resistance to maize white spot, common rust, polysora rust of maize, cercospora and leaf blights). The molecular data revealed variability among the lines, which were divided into four groups that were partially concordant with unweighted pair group method with arithmetic mean (UPMGA) and Bayesian clusters. The lines G3, G4, G11, and G13 exhibited favorable morphological characters and low disease incidence rates. The four groups were confirmed using the Gower distance in the UPGMA cluster; however, there was no association with the dissimilarity patterns obtained using the molecular data. The absence of a correlation suggests that both characterizations (morphoagronomic and molecular) are important for discriminating among elite popcorn lines.

We assessed the impact of genetic divergence and the ability to combine corn hybrids used for the production of silage on the agronomic and bromatological traits of silage quality. We evaluated 18 corn hybrids used as genitors in a circulant diallel scheme in which each genitor hybrid participated in 9 hybrid combinations, and evaluated 100 treatments [18 genitor hybrids, 81 diallelic hybrids, and a commercial check hybrid (DKB330)] in a triple lattice 10 x 10 experimental design in two environments in Brazil. Genetic variability was adequate among the corn silage hybrids, and we can recommend the use of genitors 2B688 and P30B39 for the formation of a base population for intrapopulational breeding. The P30P34 hybrid is the best for intrapopulational breeding when aiming for silage with high protein content, low fiber content, and higher in vitro digestibility. Interpopulational breeding directed at improving silage digestibility can use a combination of genitors P30P34 and AS1572, but AS1572 and P30K64 are the most recommended. Hybrids 2B688, P30P34, and SG6015 are considered the most genetically distant of the others hybrids, and have desirable combining potential; therefore, they are important genitors for the formation of new segregated populations for improving corn silage.

One of the major difficulties faced by popcorn breeders is the negative correlation between popping expansion (PE) and grain yield (GY). It is necessary to overcome this difficulty to obtain promising genotypes. One helpful tool in this process is a selection index because it allows multiple features of interest to be selected. Thus, the present study proposes a new and comprehensive selection index applied in 169 half-sib families in UEM-Co1 and UEM-Co2 composites during two cycles of recurrent selection. An experiment was conducted in a 13 x 13 lattice square in the 2004/2005 and 2006/2007 crop years in Maringá, Paraná State, and PE and GY were evaluated. To calculate Fi statistics, the following relative importance (RI) assignments were used: 0.5 for both PE and GY, and 0.70 and 0.30 for PE and GY, respectively. Families were classified according to Fi values such that Fi = 0 indicated that genotypes met the average of those selected by direct selection, Fi < 0 indicated that genotypes fell below the average of those selected, and Fi > 0 indicated that genotypes exceeded the average of those selected. Thus, desirable values of Fi were positive, indicating that the selected families were higher than those families that would be selected by direct selection for both traits. Therefore, we concluded that the novel Fi statistic was satisfactory for family selection because simultaneous and higher gains for both traits in both composites were obtained.

Increasing phosphorus use efficiency in agriculture is essential for sustainable food production. Thus, the aims of this study were: i) to identify phosphorus use efficiency (PUE) in popcorn lines during the early plant stages, ii) to study the relationship between traits correlated with PUE, and iii) to analyze genetic diversity among lines. To accomplish this, 35 popcorn lines from Universidade Estadual de Maringá breeding program were studied. The experiment was conducted in a growth chamber using a nutrient solution containing two concentrations of phosphorus (P): 2.5 µM or low P (LP) and 250 µM or high P (HP). After 13 days in the nutrient solution, root morphology traits, shoot and root dry weight, and P content of the maize seedlings were measured. A deviance analysis showed there was a high level of genetic variability. An unweighted pair group method with arithmetic mean (UPGMA) clustering analysis identified three groups for the LP treatment (efficient, intermediate, and inefficient) and three groups for the HP treatment (responsive, moderately responsive, and unresponsive). The results of a principal component analysis and selection index were consistent with the UPGMA analysis, and lines 1, 2, 13, 17, 26, and 31 were classified as PUE.

In this study, we analyzed dominant molecular markers to estimate the genetic divergence of 26 popcorn genotypes and evaluate whether using various dissimilarity coefficients with these dominant markers influences the results of cluster analysis. Fifteen random amplification of polymorphic DNA primers produced 157 amplified fragments, of which 65 were monomorphic and 92 were polymorphic. To calculate the genetic distances among the 26 genotypes, the complements of the Jaccard, Dice, and Rogers and Tanimoto similarity coefficients were used. A matrix of Dij values (dissimilarity matrix) was constructed, from which the genetic distances among genotypes were represented in a more simplified manner as a dendrogram generated using the unweighted pair-group method with arithmetic average. Clusters determined by molecular analysis generally did not group material from the same parental origin together. The largest genetic distance was between varieties 17 (UNB-2) and 18 (PA-091). In the identification of genotypes with the smallest genetic distance, the 3 coefficients showed no agreement. The 3 dissimilarity coefficients showed no major differences among their grouping patterns because agreement in determining the genotypes with large, medium, and small genetic distances was high. The largest genetic distances were observed for the Rogers and Tanimoto dissimilarity coefficient (0.74), followed by the Jaccard coefficient (0.65) and the Dice coefficient (0.48). The 3 coefficients showed similar estimations for the cophenetic correlation coefficient. Correlations among the matrices generated using the 3 coefficients were positive and had high magnitudes, reflecting strong agreement among the results obtained using the 3 evaluated dissimilarity coefficients.

The objective of the current study was to apply molecular markers (microsatellites) in the analysis of genetic diversity of 48 lines of the elite maize germplasm stored in the bank of the Cooperativa Central de Pesquisa Agrícola - Coodetec, PR, Brazil, and estimate the correlation between genetic distance and heterosis and hybrid performance from the crosses among these maize lines. Forty-four random primers were used and amplification of 124 polymorphic fragments was obtained. The expected findings from the correlation of the yield and heterosis with the genetic distance were non-significant. However, the results suggested that data from the extreme distances could be used in breeding for more productive crosses and heterotic hybrids. Thereby, molecular markers are efficient tools for predicting hybrid performance.

The purpose of this study was to analyze the genetic diversity of 15 sugary-1 sweet corn lines by microsatellite markers. One hundred pairs of simple sequence repeat primers that were mapped for field corn were tested. Of these primers, 15% were polymorphic, and all were selected for the evaluation. These primers identified a total of 39 alleles among the 15 loci that were evaluated. The number of alleles per locus in the genotypes ranged from 2 to 4, with an average of 2.60 alleles per locus; the highest number of alleles was observed at the loci Bnlg1083, Umc1241, and Umc1590. The occurrence of null alleles at locus Umc1363 was evident only in line DN44. The proportion of polymorphic loci was the highest in lines DN17.1 and DN6 (73.33%), whereas lines DN47, DN23, and DN28 were more monomorphic than other lines. The loci Bnlg1083 and Umc1506 were polymorphic in 8 and 7 lines, respectively, indicating that these loci might be effective and promising for the identification of polymorphism in other sweet corn lines. The genetic diversity calculated by Rogers' genetic distances indicated the lowest genetic similarity between lines DN9 and DN28 (0.7603) and the highest similarity between lines DN19 and DN6 (0.3724). The dendrogram obtained by the unweighted pair-group method based on arithmetic averages indicated the formation of 4 major groups, showing the crossing of the genotypes DN19 and DN6 with DN8 as a possible alternative for the expression of heterozygosis.

The fall armyworm, Spodoptera frugiperda, is the pest that causes the greatest economic losses for both common corn and popcorn crops, and the use of resistant plant genotypes is an important tool for integrated pest management. The goal of the present study was to evaluate the damage caused by S. frugiperda on single-cross popcorn hybrids under field conditions with natural infestation as well as to study the effect of 11 popcorn hybrids on the S. frugiperda life cycle under laboratory conditions. A completely randomized block design with 4 replicates was used for the field experiment, and a completely randomized design with 10 replicates was used for the laboratory experiment. In the field experiment, the damage caused by fall armyworm, grain yield, and popping expansion were quantified, and a diallel analysis was performed to select the best hybrids. For the laboratory experiment, caterpillars were obtained from laboratory cultures kept on an artificial diet and were fed with leaves from the 11 hybrids. Hybrids P7.0 x P9.4, P7.1 x P9.6, P7.2.0 x P9.3, P7.4.0 x P9.1 and P7.4.1 x P9.4 exhibited negative specific combining ability for injury by fall armyworm and positive specific combining ability for yield and popping expansion. In the laboratory experiment, the hybrids influenced the mean larval stage duration, mean larval mass, final larval mass, pupal stage duration, mean pupal mass, and adult longevity.

Corn grits are used for various purposes such as flakes, snacks, livestock feed, hominy, extruded products, beer, etc. The grit size proportion varies according to the hybrid, and thus, once the use of the grits is linked to the particle size, determining the genetic effects is essential to develop hybrids for any specific use. For this purpose a complete diallel series of crosses, involving eight parents, was performed near Maringá, PR, Brazil. The objective of this study was to evaluate the general (GCA) and specific (SCA) combining abilities of 28 progeny for selection of hybrids for breeding programs and extraction of inbred lines for hybrid development. The response variables, such as plant height, ear insertion height, crop stand, grain yield, and grits, small grits and bran production, were gauged and appraised for each of the 28 progeny. The trait effects and GCA were significant for all response variables, while for SCA, only grain yield and crop stand showed significance (P < 0.05), according to Griffing (1955) analysis. A significant weak negative partial correlation was found between grain yield and grits conversion. In relation to the hybrid selection for breeding programs, the parent IAC Nelore was highly recommended for recurrent selection and the hybrids IPR 119 x HT 392 and IAC Nelore x HD 332 for the extraction of pure lines for hybrid development.

We analyzed genetic structure and diversity among eight populations of popcorn, using SSR loci as genetic markers. Our objectives were to select SSR loci that could be used to estimate genetic diversity within popcorn populations, and to analyze the genetic structure of promising populations with high levels of heterozygosity that could be used in breeding programs. Fifty-seven alleles (3.7 alleles per locus) were detected; the highest effective number of alleles (4.21) and the highest gene diversity (0.763) were found for the Umc2226 locus. A very high level of population differentiation was found (F(ST) = 0.3664), with F(ST) for each locus ranging from 0.1029 (Umc1664) to 0.6010 (Umc2350). This analysis allowed us to identify SSR loci with high levels of heterozygosity and heterozygous varieties, which could be selected for production of inbred lines and for developing new cultivars.

Studies of genetic effects of early selection of maize based on seed quality traits are rare, especially those that use materials from different heterotic groups. These studies are also useful in tropical environments and for the advancement of sustainable agriculture with cropping during seasons not commonly used for cultivation. We estimated, through diallel crosses, the predominant genetic effects on the expression of agronomic traits and seed quality and on the general combining ability of nine maize lines from commercial hybrids and the specific combining ability of hybrid combinations among them. In the evaluation of seed quality, seven tests were used: first count and final count of seed germination, seedling vigor classification, cold tolerance, seedling emergence rate in a sand seedbed, speed of emergence in a sand seedbed, and speed of emergence index. Plant height, first ear height and grain yield were the estimated agronomic traits. In the diallel analysis, method 3 (model I) proposed by Griffing was used. There was a greater significance of non-additive genetic effects in the genetic control of seed quality of the various lines. The Flash, Dekalb 350 and P 30F80 lines combined high seed quality and high grain yield. For growth during the normal planting season, the combinations CD 3121-1 x P 30F80, Speed x CD 3121-2, Dow 8330 x AG 8080 and Dekalb 350 x CD 3121-2 were the most promising for both seed quality and agronomic traits.

Cytological analysis under light microscopy of the single hybrid P30R50 of silage corn revealed an abnormal pattern of microsporogenesis that affected the meiotic products. Meiosis progressed normally until diakinesis, but before migration to the metaphase plate, bivalents underwent total desynapsis and 20 univalent chromosomes were scattered in the cytoplasm. At this stage, meiocytes also exhibited a number of chromatin-like fragments scattered throughout the cell. Metaphase I was completely abnormal in the affected cells, and univalent chromosomes and fragments were distributed among several curved spindles. Anaphase I did not occur, and each chromosome or group of chromosomes originated a micronucleus. After this phase, an irregular cytokinesis occurred, and secondary meiocytes with several micronuclei were observed. Metaphase II and anaphase II also did not occur, and after the second cytokinesis, the genomes were fractionated into polyads, generating several unbalanced microspores, with various-sized nuclei. About 35% of the tetrads were abnormal in the hybrid. This spontaneous mutation had been previously reported in a USA maize line called ms17 and was found to cause male sterility.

We estimated the genetic diversity of 49 accessions of the hot pepper species Capsicum chinensis through analyses of 12 physicochemical traits of the fruit, eight multi-categorical variables, and with 32 RAPD primers. Data from the physicochemical traits were submitted to analysis of variance to estimate the genetic parameters, and their means were clustered by the Scott-Knott test. The matrices from the individual and combined distance were estimated by multivariate analyses before applying Tocher's optimization method. All physicochemical traits were examined for genetic variability by analysis of variance. The responses of these traits showed more contribution from genetic than from environmental factors, except the percentage of dry biomass, content of soluble solids and vitamin C level. Total capsaicin had the greatest genetic divergence. Nine clusters were formed from the quantitative data based on the generalized distance of Mahalanobis, using Tocher's method; four were formed from the multi-categorical data using the Cole-Rodgers coefficient, and eight were formed from the molecular data using the Nei and Li coefficient. The accessions were distributed into 14 groups using Tocher's method, and no significant correlation between pungency and origin was detected. Uni- and multivariate analyses permitted the identification of marked genetic diversity and fruit attributes capable of being improved through breeding programs.

In breeding programs, commercial hybrids are frequently used as a source of inbred lines to obtain new hybrids. Considering that maize production is dependent on viable gametes, the selection of populations to obtain inbred lines with high meiotic stability could contribute to the formation of new silage corn hybrids adapted to specific region. We evaluated the meiotic stability of five commercial hybrids of silage corn used in southern Brazil with conventional squashing methods. All of them showed meiotic abnormalities. Some abnormalities, such as abnormal chromosome segregation and absence of cytokinesis, occurred in all the genotypes, while others, including cytomixis and abnormal spindle orientation, were found only in some genotypes. The hybrid SG6010 had the lowest mean frequency of abnormal cells (21.27%); the highest frequency was found in the hybrid P30K64 (44.43%). However, the frequency of abnormal meiotic products was much lower in most genotypes, ranging from 7.63% in the hybrid CD304 to 43.86% in Garra. Taking into account the percentage of abnormal meiotic products and, hence, meiotic stability, only the hybrids CD304, P30K64, SG6010, and P30F53 are recommended to be retained in the breeding program to obtain inbred lines to create new hybrids.

In Brazil, using combining ability of popcorn genotypes to achieve superior hybrids has been unsuccessful because the local genotypes are all members of the same heterotic group. To overcome this constraint, 10 lines (P(1) to P(10)) with different adaptations to tropical or temperate edaphoclimatic environments were used to obtain 45 F(1) hybrids in a complete diallel. Pepper species of the genus Capsicum have been cultivated over centuries, producing both pungent and sweet fruit; the pungency is caused by alkaloids called capsaicinoids. Among the five cultivated species, Capsicum chinense is one of the most popular, being native to the Amazon basin. This species is characterized by a wide variety of fruit sizes, shapes and colors, with different capsaicinoid content. In addition, fruits are rich in vitamins A and C. Despite the importance of this plant as a spice and its medicinal uses, research on its genetic variability and potential for breeding programs is still incipient. We investigated the genetic control of some traits through diallel analysis with the objective of introgressing these traits into cultivated varieties. For the diallel analysis, the progeny of crosses between peppers with pungent and sweet fruits, together with the parents, were grown in pots under greenhouse conditions. The fruits were harvested and analyzed for the traits total fresh fruit mass, total dry fruit mass, percentage dry matter, total soluble solids, vitamin C content, fruit pungency, and number of seeds per fruit. Genetic variability was detected for all traits. In the diallel analysis, the additive-dominant model was considered to be adequate for total fresh fruit mass, percentage dry matter, total soluble solids, and vitamin C content. Additive genetic effects and dominance were found for all traits; consequently, breeding for improvement of these fruit traits would be viable.

Using only one type of marker to quantify genetic diversity generates results that have been questioned in terms of reliability, when compared to the combined use of different markers. To compare the efficiency of the use of single versus multiple markers, we quantified genetic diversity among 10 S(7) inbred popcorn lines using both RAPD and SSR markers, and we evaluated how well these two types of markers discriminated the popcorn genotypes. These popcorn genotypes: "Yellow Pearl Popcorn" (P1-1 and P1-5), "Zélia" (P1-2 and P1-4), "Curagua" (P1-3), "IAC 112" (P9-1 and P9-2), "Avati Pichinga" (P9-3 and P9-5), and "Pisankalla" (P9-4) have different soil and climate adaptations. Using RAPD marker analysis, each primer yielded bands of variable intensities that were easily detected, as well as non-specific bands, which were discarded from the analysis. The nine primers used yielded 126 bands, of which 104 were classified as polymorphic, giving an average of 11.6 polymorphisms per primer. Using SSR procedures, the number of alleles per locus ranged from two to five, giving a total of 47 alleles for the 14 SSR loci. When comparing the groups formed using SSR and RAPD markers, there were similarities in the combinations of genotypes from the same genealogy. Correlation between genetic distances obtained through RAPD and SSR markers was relatively high (0.5453), indicating that both techniques are efficient for evaluating genetic diversity in the genotypes of popcorn that we evaluated, though RAPDs yielded more polymorphisms.

We estimated general and specific combining abilities and examined resistance to northern leaf blight (Exserohilum turcicum) and to gray leaf spot (Cercospora zeae-maydis) in a set of nine inbred popcorn lines. These inbreds were crossed in a complete diallel scheme without reciprocals, which produced 36 F(1) hybrids. Two experiments with a square lattice design and three replications were conducted during the 2008/2009 crop season, in Maringá, PR, Brazil. The severity of northern leaf blight and gray leaf spot was assessed under natural infestation conditions. Data were examined by individual and joint analysis of variance. Individual and joint Griffing's diallel analyses were carried out for adjusted means. General combining ability and specific combining ability were significant (P < 0.10) by the F-test for northern leaf blight and gray leaf spot infestation levels. This denotes that additive and non-additive gene effects both contributed to resistance to these diseases, but that the additive gene effects were more important. Among the inbred lines, P(8) and P(9) gave the highest resistance to northern leaf blight, and P(3) and P(4.3) gave the highest resistance to gray leaf spot. The hybrids P(7.4) x P(8) and P(4.3) x P(9) could be exploited by reciprocal recurrent selection to provide genotypes with both northern leaf blight and gray leaf spot resistance. Significant interaction between general combining ability and crop season (P < 0.10) denotes the importance of environment, even though the disease levels in the hybrids were quite consistent.

Diallel analysis was used to obtain information on combining ability, heterosis, estimates of genetic distances by random amplified polymorphic DNA (RAPD) and on their correlations with heterosis, for the popcorn varieties RS 20, UNB2, CMS 43, CMS 42, Zélia, UEM J1, UEM M2, Beija-Flor, and Viçosa, which were crossed to obtain all possible combinations, without reciprocals. The genitors and the 36 F(1) hybrids were evaluated in field trials in Maringá during two growing seasons in a randomized complete block design with three replications. Based on the results, strategies for further studies were developed, including the construction of composites by joining varieties with high general combining ability for grain yield (UNB2 and CMS 42) with those with high general combining ability for popping expansion (Zélia, RS 20 and UEM M2). Based on the RAPD markers, UEM J1 and Zélia were the most genetically distant and RS 20 and UNB2 were the most similar. The low correlation between heterosis and genetic distances may be explained by the random dispersion of the RAPD markers, which were insufficient for the exploitation of the popcorn genome. We concluded that an association between genetic dissimilarity and heterosis based only on genetic distance is not expected without considering the effect of dominant loci.

Cocona (Solanum sessiliflorum Dunal) is a solanaceous shrub native to the Amazon region that produces an edible fruit. This species has numerous advantages, particularly a high nutritional value and productivity. However, due to irregular germination and rapid loss of seed viability, there are few plantations for production on a large scale. Development of alternative propagation strategies is essential for the production of homogeneous seedlings of genotypes with superior agronomic performance. We developed techniques for in vitro regeneration of the cocona varieties Santa Luzia and Thaís for large-scale production of healthy plantlets. Twenty days after seeding, seedling segments germinated in vitro were used as explant sources. Three successive experiments were performed: one to test the effect of the explant source and combinations of two growth regulators, auxin (indole acetic acid, IAA) and kinetin (KIN), on the morphogenetic response; another to investigate the effect of the combination of growth regulators on the morphogenetic response of hypocotyl segments, and another to evaluate how sucrose concentration affects the development of adventitious shoots. The best shoot induction was obtained using hypocotyl segments and stem apices, while rhizogenesis was greatest in leaves with a petiole. The number of adventitious shoots per explant on hypocotyl segments increased with 10 and 20 mg/L KIN, combined with 0.02 mg/L IAA in the variety Santa Luzia. Sucrose combined with these growth regulator levels increased the average number of calli; these were optimally produced when 45 g/L sucrose and 0.01 mg/L IAA + 20 mg/L KIN were applied. Only sucrose concentration influenced shoot proliferation in the two S. sessiliflorum varieties, with a maximum at 17.5 g/L.
